摘要
胃蛋白酶消化牛腱胶原的提取纯化以及作为BMP的载体复合后注射入小鼠肌肉。术后7天组织学检查可见异位成骨,并有新生血管,胶原部分吸收。28天时软骨被吸收并被有红骨髓的正常骨组织替代,大部分胶原被吸收。未见排斥反应。钙含量测定显示成骨量大于单纯植入BMP组。单纯注射胶原组未见成骨。实验结果表明牛腱胶原经胃蛋白酶消化等提取纯化处理后,抗原性很弱,对部分纯化的BMP有助溶助悬作用,吸收快,利于成骨,是BMP的一种良好缓释载体。该种胶原与BMP的复合物具有较大临床价值。
Pepsindigestedbovinetendoncolagenwaspurifiedandinjectedasasustainedreleasecarrierforbonemorphogeneticprotein(BMP)inthethighofthemouse.Histologicexamination,at7dayspost-implantation,showedectopicboneformationandcapilaryformation.Andthecolagenwasabsorbedpartialy.Thecartilagewasresorbedandreplacedbynormalbonecontaininghematopoieticbonemarrow28daysafterimplantationandthecolagenalmostcompletelydisappearedatthesametime.Noimmunerejectionwasseen.CalciumcontentmeasurementindicatedmorenewboneformationthantheBMPalone.TheseresultsdemonstratedthatthecolagenisefectiveasacarrierofBMP,withtheadvantageofkeepingthelaterstableinthecolagensolution,enhancingthenewboneyieldinduc-tionbyBMPandbeingrapidlybiodegradedbyhosttissues.Apparentlythiscompostitemaybeofvalueinorthopaedicsurgery.
