非霍奇金淋巴瘤病人外周血T细胞TCR Vβ谱系的选择性利用和克隆性增殖分析 被引量:3

摘要 为了解B细胞性非霍奇金淋巴瘤 (B NHL)和T细胞性非霍奇金淋巴瘤 (T NHL)病人TCRVβ亚家族T细胞的分布及其克隆性的情况 ,利用RT PCR分别扩增 4例B NHL和 2例T NHL病人外周血单个核细胞的TCRVβ 2 4个亚家族基因的CDR3,了解病人各Vβ亚家族的利用情况。阳性的PCR产物进一步经荧光素标记和基因扫描分析产物的CDR3长度 ,以了解T细胞克隆性。结果显示 ,6例NHL病人外周血T细胞仅选择性表达 6 - 12个Vβ亚家族 ,全部病人均选择了表达Vβ1,Vβ8,Vβ13和Vβ19,5例病人表达Vβ2和Vβ16 ,而全部病人均未能检测到Vβ4 - 6 ,Vβ10 - 12 ,Vβ15 ,Vβ17- 18,Vβ2 0 ,Vβ2 2 - 2 3。基因扫描发现 2例B NHL和 1例T NHL病人的 1- 3个Vβ亚家族T细胞呈克隆性增殖。结论提示 ,B NHL和T NHL病人外周血T细胞具有相似的Vβ亚家族选择性利用的特点 。 To investigate the distribution and clonal expansion of TCR Vβ subfamily T cells in patients with B NHL and T NHL, the CDR3 of TCR Vβ 24 subfamily genes was amplified in peripheral blood mononuclear cells from 4 cases with B NHL and 2 cases with T NHL using RT PCR, and to observe the usage of TCR Vβ repertoire, the PCR products were further labeled with fluorescence and analyzed by genescan technique for the CDR3 size, to evaluating clonality of the detectable TCR Vβ T cells. The results indicated that only selected expression of 6-12 Vβ subfamily T cells could be identified in the 6 cases with NHL, and Vβ1, Vβ8, Vβ13 and Vβ19 were expressed in all samples, Vβ2 and Vβ16 could be found in 5 samples, whereas Vβ4-6, Vβ10-12, Vβ15, Vβ17-18, Vβ20 and Vβ22-23 were absent in all samples. Genescan analysis showed that clonal expansion of T cells could be found in 1-3 Vβ subfamilies from 2 cases with B NHL and 1 case with T NHL. In conclusions, the similar selected usage of TCR Vβ subfamily T cells could be found in peripheral blood from patients with B and T NHL, clonal expansion of T cells which were considered to be related to lymphoma cell antigen could be detected in a part of patients with B or T NHL.
