健康人服用黄芪当归合剂后体内阿魏酸的HPLC法测定 被引量:7

HPLC in Determination of Ferulic Acid in Healthy Persons after Taking Mixture of Radix Astragali seu Hedysari and Radix Angelicae Sinensis

摘要 目的 建立健康人服用黄芪当归合剂后的血清预处理方法。方法 应用沸水浴一次处理、甲醇二次处理去除血清蛋白质方法 ,用HPLC直接测定健康人服用黄芪当归合剂后体内阿魏酸 (FA)。色谱条件 :甲醇 冰醋酸 水 (4 0∶0 .3∶5 9 .7)为流动相 ,C1 8(Diamonsil)柱 (15 0mm× 4.6mm ,5 μm)为固定相。FA用内标 (香豆精 )法定量。 结果 FA的最低检测限为 2 .5 1ng ;沸水浴 10min的最低检测血清浓度为 2 5 .10ng/mL ,线形范围 40 .16~ 80 3 2 .0 0ng ,r=0 .9975。黄芪当归合剂口服后 5min血清中即能测出阿魏酸 ,在 3 0min达高峰 ,在 90min出现第 2次高峰。结论 该方法保留了水浴法的优点 ,灵敏、快速、简便。 OBJECTIVE To establish the method of pre-treatment of serum of healthy persons after taking Mixture of Radix Astragali seu Hedysari and Radix Angelicae Sinensis .MEHTOD With boiling water bath method used in the primary treatment and methanol in the secondary treatment to remove serum proteins, HPLC was employed to directly determine the content of ferulic acid in healthy persons after taking Mixture of Radix Astragali seu Hedysari and Radix Angelicae Sinensis . Colour spectrum condition: methanol glacial acetic acid-water (40∶0.3∶59.7) as the mobile phase, C 18 (Diamonsil) column (150 mm×4.6 mm, 5 μm) as the stationary phase. Ferulic acid was quantified with internal standard method (coumarin). RESULT The minimum determination limit of ferulic acid was 2.51 ng; the minimum serum concentration in ten minute boiling water bath detection was 25.10 ng/mL; linear range was 40.16 ~ 8 032.00 ng; r =0.997 5. Five minutes after taking the mixture, ferulic acid was detected in the serum, and the first and second peaks appeared 30 minutes and 90 minutes after the intake of the mixture. CONCLUSION This method maintains the advantages of water bath method, with flexibility, swiftness, simplicity, specificity and good repeatability.
