摘要
目的:建立高效液相色谱串联质谱测定家兔血样中阿魏酸的含量测定方法。方法:采用高效液相色谱串联四级杆质谱检测血样中阿魏酸的含量,采用电喷雾(ESI)离子源,负离子扫描,二级反应监测模式(SRM)进行检测。色谱柱为ODSC_(18)柱(4.6 mm×250 mm,5 μm);流动相为甲醇;流速为0.25 ml·min^(-1)。血浆样品经乙醚萃取,高速离心后进样分析;监测离子为:母离子m/z 193.03^(-1),子离子m/zl 33.9。结果:阿魏酸在5.6~280μg·L^(-1)范围内线性关系良好,最低定量限为5.6 μg·L^(-1),r=0.9991。阿魏酸的日内和日间精密度RSD均小于5%,绝对回收率>86%。结论:高效液相色谱串联质谱测定血样中阿魏酸的定性定量分析方法,专属性强、快速、灵敏、可靠,可满足以阿魏酸为主要成分的药物的药动学研究的需要。
OBJECTIVE To establish an HPLC-MS MS method for the determination of ferulic acid of ordinary Guipiwan and uhra-micro Guipiwan in rabbit. METHODS HPLC-MSMS was used to identify the concentration of the ferulic acid in the blood by using ionization source of ESI in negative ion mode and detected by SRM mode. RESULTS The concentration of FA between 5.6-280μg·L^-1 had a good linear relationship, and the lowest detected limits was 5.6 μg.L^-1 ,R =0.999 1. CONCLUSION The method of identifying the concentration of the ferulic acid in the blood by HPLC-MS/MS provided an accurate, proper, reliable and fast method to identify the FA, and provided more scientific information for quality control of traditional medicine.
