摘要
作者采用高效液相色谱法同时测定多种抗癞痫药物在人血浆中的浓度。测定药物有朴痫酮、苯巴比妥、苯妥英、卡马西平和10.11一环氧化卡马西平。用于研究抗癫痫药物配伍用药后体内各药物动力学的变化及临床治疗药物的监测工作。色谱条件:ODS-C18柱,紫外检测波长210nm,流动相为甲醇和水的等体积混合溶液,流速为1.0ml/min,内标为苯乙酮。本文采用相对校正因子来控制多批样本测定质量,用t检验判断药物测定的稳定性,证明本法稳定,重复性好,灵敏度高等优点。
We established that HPLC for determinating the concentration of primidone (PM), phenobarbital (PB), phenytoin (DPH), carbamazepine (CBZ) and its metabolite carbamazepine-epoxide (ECBZ) in human plasma is a rapid and specific micro-technique. The procedure involves reverse-phase chromatography on a 4.6×250mm column containing ODS-C18. The antiepileptic drugs were eluted with en equivolume mixture of distilled water and methy-alcohol at a flow rate of 1.0ml/min, detected by their absorbance at 210nm and quantitated by measuring peak area. By taking known amounts of the five drugs and internal standard through the procedure we demonstrated a linear relationship between peak area and drug concentration. The concentration range far exceeded that required for clinical purpose. In addition, we established the relative response factors (RRF) for use in final calculation. The RRF are 1.00 for PM, 1.36 for PB, 0.51 for DPH, 0.26 for CBZ and 0.19 for ECBZ. Analytical recoveries for the five drugs varied from 95.62 to 104.01% with good precision (coefficients of variation between 3.12 and 8.85 for therapeutic and toxic concetration).
