摘要
本文采用简便的HPLC条件和萃取方法,12min内同时测定微量血清(100ul)中的苯巴比妥、苯妥英和卡马西平的含量.以苯乙酮为内标,样品用三氯甲烷萃取,N260℃吹干.流动相为McOH/H2O(55/45,V/V),流速1.0ml/min,于220nm检测.在7个不同水平,PBT、PHT、CBZ的平均回收率为99.39%、98.35%和99.69%,天内和天间变异均低于5%,在分析范围内有良好的线性关系.应用本法测定了8名病人血清中的药物浓度.
A simple,sensitive and rapid RP-HPLC method for the simultaneous deter mination of concentation of Phenobarbital (PBT),Phenytoin(PHT)and Carbamazepine(CBZ) in hUman serum is established. Using acetophenoen as internal standard and chloroform to extract drugs in plasma. The mobile phase consists of methanol - water (55/45, v/v). and flow rate is 1. 0ml/min, UV detecor 220nm. The recoveries of PBT, PHT and CBZ is 99.39%, 98. 35%and 99- 69% respectively. Within -runCV and between - run CV is less than 5%,the method has a good linearity. The assay has been used to determine the drug level in plasma in eight patients.
