食管癌HLA-A,B,C基因抗原表达下调机理的研究 被引量:3

摘要 目的 :研究食管癌及食管上皮人类白细胞抗原 (HLA A ,B ,C)的抗原表达及探索该抗原丢失的分子机理。方法 :采用抗HLA A ,B ,C单抗 ,以免疫组化法及甲基化PCR检测HLA A ,B ,C抗原在食管癌及食管上皮的分布及HLA A ,B ,C基因甲基化状况。结果 :31例食管癌中 2 7例无HLA A ,B ,C抗原表达 ,正常及异常增生的食管上皮均可见该抗原的阳性表达 ;45 2 %的食管癌HLA A ,B ,C基因甲基化 ,食管上皮该基因无甲基化。结论 :食管癌中HLA A ,B ,C抗原大部分丢失 。 Objective: Our aims were to measure the expressions of human leucocyte antigen(HLA A, B, C) antigens in esophageal squamous cell carcinomas (ESCC) and neighboring normal epithelia and to study the molecular mechanisms of the loss of their expressions. Methods: The HLA A, B, C antigens were detected by monoclonal antibody against HLA A, B, C antigens using immunohistochemistry. Methylation of the genes encoding HLA A, B, C antigens was determined by the presence of the methylated specific PCR products. Results: The HLA A, B, C antigens were absent in 27 cases of 31 tumor samples, while positive staining was observed in all samples of normal esophageal epithelia and esophageal epithelia with dysplasia.The rate of methylation of HLA A, B, C genes in ESCC was 45.2%. No methylation was found in normal esophageal epithelia. Conclusion: Loss of expression of HLA A, B, C antigens is frequent in ESCC. Methylation of HLA A, B, C gene may be one of the major mechanisms for the down regulation of their expression in ESCC.
