摘要
用鼠抗人HLA-A,B,C单抗W6/32及HLA-DR单抗HB55与细胞因子基因修饰肿瘤细胞结合,以FITC-免抗鼠Ig为二抗,间接免疫荧光法流式细胞仪测定了HLA-Ⅰ、Ⅱ类抗原表达,与未修饰及空白载体修饰肿瘤细胞相比,基因修饰肿瘤细胞的HLAI-Ⅰ、Ⅱ类抗原表达有明显增加,其中IPN-γ作用最强,分别将基因修饰及未修饰肿瘤细胞接3×106细胞/200μl/只接种于Balb/C棵鼠右前肢皮下,逐周观察接种肿瘤细胞的生长。结果表明,细胞因子基因修饰肿瘤细胞的致瘤性都有显著下降甚至消失,这为“细胞因子基因工程疫苗”的临床应用提供了一定的依据。
Indirect immunoflurescence was performed in a FACScan using monocloned antibodies W6/32 (anti HLA-A,B,C), HB55 (anti HLA-DR), and fluorescein isothiocyanate(FITC)-labled rabbit anti mouse immunoglobulin as second antibodies. Mean fluorescence intensity was given as channel number on log scale from 1 to 1 024. The percentage of cells expressing HLA-antigans was calculated upon subtraction of the respactive background fluorescence determined for each group with control.To prepare inocula, cells were harvested from stock cultures, washed twice with PBS and resuspended in PBS, 3 x 106cells/200μl were injeted s.c. into the right axilla of the Balb/c nude mice. All mice were examined once a week, and the volume of tumors was calculated according to a x b2 X 0.4, where a is the larger and b is the smaller diameter(cm). The weight of tumor tissue was gained from killed mice after 4~6weeks growth.The results show that the immunogenicity of gene-modified tumor cells was increased, while the tumorigenicity was decreased or abrogated.
