摘要
目的 观察大豆异黄酮对YAC 1细胞及小鼠ESC艾氏肉瘤生长的抑制作用及对免疫功能的影响。方法 用MTT法测定大豆异黄酮对YAC 1细胞生长的抑制率和小鼠淋巴细胞转化刺激指数 ;LDH释放法测定小鼠NK细胞活性 ;称重观察肿瘤生长和脾指数和胸腺指数。结果 金雀异黄素和大豆甙元剂量依赖性抑制YAC 1细胞生长 ,并随时间延长作用增强 ;大豆异黄酮 5 0、10 0、4 0 0mg·kg- 1灌胃给药均明显升高雌性小鼠脾指数和淋巴细胞转化的刺激指数 ,对雄性小鼠则无明显作用 ;大豆异黄酮10 0、4 0 0mg·kg- 1明显抑制小鼠ESC艾氏肉瘤生长 ,同时 5 0、10 0、4 0 0mg·kg- 1均降低异常增高的脾指数 ,增加NK细胞活性。结论 大豆异黄酮明显抑制YAC 1细胞和小鼠ESC艾氏肉瘤的生长 ,同时增强非特异性免疫功能 ,并可增强正常雌性小鼠免疫功能。
Aim To observe the effect of isoflavones on the growth of YAC-1 cells in vitro and ESC tumor in mice and their immune function. Methods Isoflavones induced inhibitory rate of YAC-1 cells and the proliferation of lymphocyte were assessed by the modified MTT assay. NK cell activities were determined by LDH. The antitumor activity in vivo was observed by weighing the tumor and calculating spleen and thymus indexes. Results The growth of YAC-1 cells were inhibited by isoflavones in a concentration and time-dependent manner. Isoflavones power extracted from soybean (50, 100, 400 mg·kg -1, oral administration) could enhance spleen index and ConA-induced proliferation of lymphocyte in female mice, but not in male ones. Isoflavones in the doses of 50, 100, 400 mg·kg -1 could significantly inhibit spleen index and increase NK cell activities. The growth of tumor was significantly inhibited in ESC tumor-bearing female mice in the doses of 100, 400 mg·kg -1. Conclusion Isoflavones has inhibitory effect on the growths YAC-1 cells in vitro and ESC tumor in vivo. Isoflavones not only strengthens the nonspecail immune function of tumor-bearing mice, but also of the normal female mice.
