摘要
目的 建立大鼠脑冷冻伤模型并探讨脑组织细胞冷冻伤中Bcl- 2的表达 ,提供抗损伤治疗的新思路 .方法 运用液氮冷冻器作用于大鼠左侧顶部脑组织后 2h、6h、12h ,建立脑冷冻伤模型 ,同时观察冷冻伤侧脑组织病理形态学改变与Bcl- 2表达的变化 .结果 液氮作用后 2h ,脑组织已出现充血、水肿及神经元坏死的表现 ,并于 12h明显累及海马回神经元 .有价值的是 ,与坏死交界处的健在神经元呈现了Bcl- 2的高表达 .结论 说明了脑组织中神经元的损伤与Bcl- 2基因蛋白的表达有关 ,提示通过抗细胞凋亡的途径对脑损伤治疗具有应用前景 .
Objective To establish rat model of brain cold injury and study on Bcl-2 protein expression and provide a new clue to protest against brain injury. Methods Left parietal side of the rat skull was subjected directly to cold brain injury resulted from liquid nitrogen freezer at the temperature of -196℃ for 1 minute. Then, the pathomorphological changes of the ipsi-lateral brain and its relationship with the expression of Bcl-2 protein were observed. Results The results were that, 2h after treatment with liquid nitrogen, the ipsi-lateral brain showed congestion, edema, and minor focal necrosis. The degeneration and necrosis of neurons aggravated as a function of time interval, with severe pathological changes of neurons at 6h and obvious expansion of hippocampus at 12h.Correspondingly,the expression of Bcl-2 protein in injured tissue, especially on the boundary of the necrosis region, and showed highest level at 12h after cold injury. Conclusions The findings suggested that,in the rat model of brain cold injury, the expression of Bcl-2 protein was closely related to the extent of neuron injury.
