摘要
对T .glabrataWSH -IP12进行EMS诱变 ,挑选以NH4 Cl为唯一氮源的平板上透明圈较大的菌株 ,经初筛和复筛后 ,发现T .glabrataWSH -IP30 3生产丙酮酸的能力强且稳定。以NH4 Cl为唯一氮源摇瓶培养 4 8h ,其丙酮酸产量 (35 .1g/L)比出发菌株 (2 1.4 g/L)提高了 64%。采用该菌株在 30 0L罐上进行了 4批发酵试验 ,丙酮酸产量最高可达 5 8.4g/L ,对葡萄糖产率0 .5 62 g/ g。
Mutants which exhibited large transparent rings in the plate with NH 4Cl as a sole nitrogen source, were obtained after ethyl methane sulfonate (EMS) treatment of T. glabrata WSH-IP12. Among them, a mutant WSH-IP303, not only produced pyruvate to a high level with NH 4Cl as a sole nitrogen source, but also showed strong genetic stability during the generation. In a defined medium containing glucose, NH 4Cl, salts and vitamins, pyruvate production by T. glabrata WSH-IP303 reached 35.1 g/L by 48 h flask culture, which was 64% higher than the parent strain. Pyruvate concentration and yield reached 58.4 g/L and 0.562 g/g, respectively, in a 300 litre pilot-scale fermentor.
