摘要
目的 :研究 Bax和 Bcl 2蛋白在放射复合伤口愈合中的表达及其与细胞凋亡的关系。方法 :用原位末端标记 (TU NEL)法检测细胞凋亡 ,用免疫组织化学方法检测 Bax、Bcl 2蛋白表达。结果 :(1)与单伤组比较 ,照射后细胞凋亡的变化具有出现较早、频度较高、消失推迟 3个显著特点 ,可能是导致辐射延迟伤口愈合的重要原因。 (2 )观察伤口愈合过程中 Bax、Bcl 2蛋白表达及其与细胞凋亡的关系 ,发现随着细胞凋亡的增加 ,Bax蛋白的表达明显增强 ,并且与细胞凋亡发生频度具有良好的相应关系。而 Bcl 2蛋白水平在细胞凋亡达高峰时呈明显下降趋势 ;当凋亡明显下降时 ,Bcl 2蛋白表达增加 ,并逐渐恢复其正常水平。结论 :Bax和 Bcl
Objective:To study the relationship between the expressions of Bax and Bcl2 proteins and apoptosis in radiation combined wound of rats.Methods:Apoptosis and Bax and Bcl2 proteins were estimated by in situ terminal labeling (TUNEL) and immunohistochemical methods.Results:(1)It was shown that apoptosis manifested three typical characteristics,namely early occurrence,high frequency and delayed disappearance after radiation,when compared with those of single wound group.This characteristics might explain the delayed wound healing caused by radiation.(2) The expression of Bax protein increased evidently with the increment of apoptosis,and it showed a good correlation with apoptosis in the process of wound healing.The expression of Bcl2 protein decreased obviously as the apoptosis reached the beak,and it showed an ascending tendency up to normal level when the apoptosis decreased distinctively.Conclusions:Bax and Bcl2 proteins played an important role in the regulation of apoptosis in the process of healing of combined wound in rats.
