摘要
本工作观察到短时间无钙液灌流的心脏恢复钙灌流时发生严重的钙反常损伤,而在复钙液内加入10^(-5)mol/LMT,显著减轻钙反常损伤。预先注射小剂量ZnSO_4诱导内源性MT合成,显著增强心脏对钙反常损伤的抵抗能力。
The effects of metallothionein(MT), which was exogenously given into the. perfusate or induced in vivo with administration of ZnSO4,on Ca paradox were, observed on isolatd perfused rat hearts in order to study the pathophy-siologic significance of its cell protective action.During reperfusion with Krebs-Henseleit(KH) buffer(Ca2 + ,1.5mmol/L) following 10 min of perfusion with Ca-free KH buffer,There showed severe damage of the hearts-myocardium contracture,steeple increases of intraventricular pressure and coronary perfusion pressure,and gain of myocardial calcium and myocardial lipid peroxide, MDA. However, reperfusion with Ca-rich KH buffer containing 10-5mol/L of MT significantly alleviated the Ca paradox-induced myocardial damage. Induction of MT synthesis in the body with preinjection of small dose ZnS04 obviously enhanced the resistance of the heart to Ca paradox.The results suggest that MT as an endogenous anti-injury substance,its synthetic form may be used for myocardial ischernia-reperfusion injury in the clinics.
