摘要
目的 :分析颌骨骨折愈合过程中总纤维粘连蛋白 (t FN)和细胞型纤维粘连蛋白 (c FN)的表达和变化 ,探讨纤维粘连蛋白在颌骨骨折愈合过程中的作用。方法 :以兔作为实验动物 ,制作下颌骨骨折动物模型 ,于骨折愈合的不同时期定期处死动物 ,获取标本 ,采用 L s AB免疫组化染色法检测 t FN和 c FN的表达 ,而同期 t FN和 c FN表达强度的差异则体现了 p FN的表达水平。结果 :血浆型纤维粘连蛋 (p FN)在正常组织中有表达 ,骨折后 1d表达明显增强 ,5~ 7d时达峰值 ,骨折后 3月在骨基质中仍有较强表达。 c FN在骨折后 3d出现表达 ,7d时达峰值 ,2周后开始减弱 ,1月时仅可观察到微弱表达 ,表达部位主要在骨周结缔组织基质。结论 :颌骨骨折愈合过程中 ,p FN与 c FN的表达存在着时空差异 ,提示 p FN主要在血凝块形成、单核细胞吞噬作用、骨基质沉积、钙化等过程中发挥作用 ,c FN对细胞分化、增殖有着重要调节作用。
砄bjective:To analyse the expression of total fibronectin(tFN) and cellular fibronectin(cFN) in fractured mandibular bone during the healing course.Methods:The rabbit mandibular bone fracture model was made,and specimens were acquired at various healing stages.The expression of tFN and cFN was determined by LsAB immunohistochemsitry.The difference between tFN and cFN was used as the real expression of plasma fibronectin (pFN).Results:pFN was detected in normal tissue,increased obviously 1d after bone fracture,and reached peak value in 5~7 d.3 months later,its expression in bone matrix still maintained at a relatively high level.cFN was detected 3 d after bone fracture,reached peak value in 7 d,decreased in 2 weeks. 1 month later only faint signal could be detected.cFN distinctly expressed in periosteal connective tissue matrix.Conclusion:The different expression of pFN and cFN may suggest that pFN and cFN have different functions during healing course of bone fracture.
