摘要
目的:探索下颌骨骨折愈合过程中纤维粘连蛋白mRNA的表达规律。方法:制作下颌骨骨折动物模型,对骨折愈合过程中不同时期的组织标本切片行mRNA原位杂交实验。结果:正常骨组织中没有检测到纤维粘连蛋白fibronectin,FN)的mRNA表达。骨折愈合过程中,在成纤维细胞、纤维细胞、血管内皮细胞、软骨细胞、破骨细胞和一些骨组织细胞中观察到FN的mRNA表达。表达的时间规律为骨折后3d出现明确阳性表达,逐步增强,7~14d达到峰值,其后急剧下降;至1月时,仅有微弱信号存在,2月后未再观察到阳性信号。结论:在下颌骨骨折愈合过程中,有多种类型的局部组织细胞出现了FNmRNA表达,合成的内源性细胞型纤维粘连蛋白(cellular fibronectin,cFN)对骨折愈合具有重要影响。
Objective: To investigate the expression of FN mRNA during mandibular fracture healing course. Methods: Rabbit mandibular bone fracture model was established, histologic specimen were obtained at different healing stages. The expression of FN mRNA was determined by in situ hybridization. Result: FN mRNA expressed obviously in fibroblasts and fibrocytes, it was also detected in some other cell types, such as vasal endothelial cells, chondrocytes, osteoclasts and some bone tissue cells. No FN mRNA signal was detected until the 3rd day after bone fracture, then the amounts increased, and reached peak value at 7-14 days. Expressional intensity decreased significantly 2 weeks later. Only faint signal was detected 1 month later. Conclusion: FN mRNA expresses in many topical cells during the healing process of bone fracture.
