牛血浆纤维粘连蛋白对体外培养的大鼠成骨细胞增殖的影响 被引量:3

摘要 目的研究外源性牛血浆纤维粘连蛋白对体外培养的大鼠成骨细胞增殖的影响。方法采用体外培养技术,原代培养大鼠成骨细胞,用AlamarBlueTM摄入法,检测不同浓度的牛血浆纤维粘连蛋白对大鼠成骨细胞增殖的影响。结果体外培养的成骨细胞呈梭形、三角形或多角形,多角形细胞有多个矢状突起;核卵圆形,常居一侧;胞质内碱性磷酸酶染色呈阳性反应,Ⅰ型胶原免疫组化染色呈阳性。牛血浆纤维粘连蛋白能够促进大鼠成骨细胞的增殖,当浓度为40μg/ml时,AlamarBlueTM还原率明显升高,与对照组相比有显著性差异(P<0.05);当浓度为50μg/ml时,差异极显著(P<0.01)。结论一定浓度的牛血浆纤维粘连蛋白具有促进大鼠成骨细胞增殖的作用。 Objective To investigate the effects of bovine plasma fibronectin on the proliferation of rat osteoblast in vitro.Methods Cell cultured.AlamarBlueTMwere used to determine the effect of various concentration of bovine plasma fibronectin on the proliferation of rat osteoblast in vitro.Results Fibronectin can stimulate the proliferation of rat os-teoblasts.Statistically significant difference was detected at dose of40μg/ml(P<0.05)and dose of50μg/ml(P<0.01).Con-clusions Certain concentration of bovine plasma fibronectin can stimulate the proliferation of rat osteoblast in vitro.
