应用抑制性消减杂交分析全反式维甲酸和1,25-二羟维生素D_3诱导结肠癌细胞的相关基因 被引量:6

Analysis of the Differentiation Associated Genes in Colon Cancer Cells Induced by All-trans-retinoic acid and 1,25- Dihydroxyvitamin D_3 Using SSH Technology

摘要 为探讨结肠癌细胞诱导分化的机制 ,采用抑制性消减杂交 (suppression subtractivehybridization,SSH)研究联合使用全反式维甲酸和 1 ,2 5-二羟维生素 D3诱导分化结肠癌 Lo Vo细胞前、后差异表达的基因 .经比较消减 c DNA文库的序列与基因库的序列 ,发现 :有 1个基因的序列与正常鳞状上皮细胞中的 1个表达序列标签 (expressed sequence tag,EST)高度同源 ,同时发现6个新 EST (基因库登录号为 AW2 66492、AW2 66493、AW2 66494、AW58751 8、AW58751 9和AW58752 0 ) .说明诱导分化涉及到多个基因的表达 ,结肠癌的发生是多基因综合作用的结果 .进一步研究这些基因和 EST的功能对于结肠癌的防治将有重要意义 . Inducing differentiation on tumors is termed as the re\|differentiation of malignant tumor cells to normal cells by some of internal and extrasomatic inducers.All\|\%trans\%\|retinoic acid (ATRA),a derivative of vitamin A,is known as the most common inducer.1,25\|dihydroxyvitamin D\-3 also has a good potential for treating tumors.It is necessary to continuously study the molecular mechanism of inducing differentiation.The difference of gene expression in colon cancer cells before and after combinative use of ATRA and 1,25\|(OH)\-2D\-3 was screened by suppression subtractive hybridization (SSH) technology.Then the subtracted cDNA library was established and its sequence was compared with GenBank.The results demonstrated that a sequence was highly homologous with an expressed sequence tag(EST) from normal squamous epithelium cell and 6 new ESTs were accessed by GenBank.Their numbers are AW266492,AW266493,AW266494,AW587518,AW587519 and AW587520.Since many ESTs have been found in this inducing differentiation,it means that many genes take part in the inducing differentiation.To continuously study the functions of those ESTs will be useful for the treatment and prevention on colon cancer.
