摘要
目的研究上海地区血压正常汉族老年人中(年龄≥ 60岁) I型血管紧张素 II受体( AT1 R)基因 A1166C多态性的分布特点,并同国内外人群的分布频率相比较,了解地区种族分布差异及与年龄、体重指数 BMI( kg/m2)的相互关系。方法体检选取 92例正常健康老年人,男 72例,女 20例,平均年龄 65.32± 4.70岁。应用 PCR RFLP方法检测位于 AT1 R基因 3' UTR的 A1166C变异。结果( 1)上海地区血压正常汉族老年人 AA、 AC、 CC基因型频率分别为 0.89、 0. 11、 0. 00, 1166C等位基因频率为 0. 05。低于白种人和日本人( P<0.001和 P=0.046),与美国黑人相比无差异 (P=0.815)。( 2)在国内人群中 C1166等位基因的分布频率无差异,并与年龄、体重指数无关。结论 AT1 R基因 A1166C多态性存在种族地域间的差异,上海地区与国内现有报道结果一致, 1166C等位基因频率明显低于白种人群,但不同年龄不同体重指数人群 1166C等位基因频率无差别。提示由于中国人群 1166C等位基因频率低,故开展 AT1 R基因 A1166C多态性与疾病的关系中需要大样本研究。
Objective To observe the distribution of A1166C polymorphism of the type I angiotensin II receptor gene in elderly Chinese Han people (≥ 60 years )who have normal blood pressure,and to compare the allele frequency of 1166C in Chinese group and that in alien group,so as to find the racial and regional distribution difference and its association with age and BMI (kg/m2).Methods The population study comprised of 92 normotensive subjects (male=72,female=20)from Shanghai whose average age was 65.32± 4.70.A→ C polymorphic site at 1166 nucleotide position in the 3' untranslated region of AT1 R gene was determined by using restriction fragment length polymorphism analysis of DNA generated by polymerase chain reaction (PCR RFLP). Results (1)The frequencies of AA、 AC、 CC genotype in the elderly Shanghai Han people were 0.89,0.11and 0 respectively .The 1166C allele frequency was 0.05.In present report that allele frequency of 1166C in normotensive Chinese Han people was lower than that in Whites and Japanese (P< 0.001and P=0.046),but no difference was found in comparison with Africans Amercians (P=0.815).(2)The 1166C allele frequency had no association with age and BMI in elderly Chinese Han people.Conclusions Because of the lower frequency of the AT1 R 1166C allele in elderly Chinese Han people,the detection of a significant association of AT1 R 1166C with hypertension would require a large sample size.
