摘要
目的 :探讨细胞表面糖基在胚泡着床中的作用。方法 :用刀豆凝集素 (ConA)、荆豆凝集素(UEA)、麦胚凝集素 (WGA)、花生凝集素 (PNA)和麦胚凝集素 (WGA)为分子探针作亲合细胞化学染色 ,检测了与 4种凝集素相对应的受体糖基在小鼠子宫内膜中的表达变化。结果 :在胚泡植入期间 ,ConA受体一直为弱阳性 ,变化不明显 ;UEA受体主要分布于内膜表面上皮和腺上皮细胞表面 ,随妊娠天数增加含量逐渐减少直至消失 ;PNA受体出现于孕 5d ,在着床处的蜕膜细胞膜上明显增多 ;着床前期 ,WGA受体主要分布于子宫内膜表面上皮和腺上皮细胞膜上 ,胚胎植入后 ,WGA受体在着床处的蜕膜细胞膜上明显增多。结论 :在着床期子宫内膜各种细胞的凝集素受体糖基呈现规律性变化 ,提示子宫内膜糖基的变化与胚泡着床密切相关。
Objective:To observe the changes of the oligosaccharides on cell surface in the process of implantation.Methods:Four different biotinylated lectins[ConA(mannose specific),UEA(fucose specific),PNA(galactose specific),WGA(sialic specific)] were used as molecular probes to observe the distribution and quantity of related oligosaccharides in the endometrium during early pregnancy.Results:ConA showed weak staining in the endometrium;UEA band to the cell surface of luminal and glandual epithelium at the begining of implantation,then decreased and losed.The staining reaction of PNA were present at day 5 of prenancy,and increased in the surface of decidual cells at implantation site.The binding of WGA distributed throughout the endometrium,with intense staining around the implantation site.Conclusion:The changes of oligosaccharides imply the glicoconjugates,including sialic acid,galactose and fucose may play important roles during implantation.
