摘要
目的:建立HPLC同时测定栝楼桂枝汤中没食子酸、原儿茶酸、芍药内酯苷、芍药苷、肉桂酸、桂皮醛、甘草苷、甘草素、甘草酸含量的方法,并探讨君药天花粉对复方9种成分的含量影响。方法:用Diamonsil C18柱,流动相0.1%甲酸-乙腈梯度洗脱,流速0.8 mL·min-1,柱温30℃,检测波长254 nm;测定桂枝汤、栝楼桂枝汤中没食子酸、原儿茶酸、芍药内酯苷、芍药苷、肉桂酸、桂皮醛、甘草苷、甘草素、甘草酸的含量,比较了配伍天花粉的栝楼桂枝汤与桂枝汤9种成分的含量变化。结果:建立了HPLC同时测定没食子酸、原儿茶酸、芍药内酯苷、芍药苷、肉桂酸、桂皮醛、甘草苷、甘草素、甘草酸的含量的方法,与桂枝汤相比,栝楼桂枝汤中芍药苷含量增加、没食子酸、原儿茶酸、芍药内酯苷、甘草苷、甘草素、肉桂酸、甘草酸含量减少,桂皮醛含量变化不大。结论:本方法专属性、重复性好,可用于栝楼桂枝汤中9种成分的含量测定;配伍天花粉对各成分含量有影响,其影响非常复杂。
Objective: To establish an HPLC method for the determination of the nine active ingredients( gallic acid, protocatechuic acid, albiflorin, paeoniflorin, glycyrrhizin, liquiritin, cinnamic acid, cinnamaldehyde, glycyrrhizic acid) of Gualou Guizhi decoction (GLGZD) and Guizhi decoction (GZD), and to determine the change of general contents in different decoctions. Method: The chromatographic separation was performed on C]s (4.6 mm ×250 mm, 5 μm) nalytical column with mobile phase A-aeetonitrile, B-O. 1% formic acid at a flow rate of O. 8 mL·min-1. The column temperature was at 30 ℃ ; the detection wavelength was set at 254 nm. The contents of the above-mentioned nine constituents were determitaed in GZD and GLGZD, respectively. Result: The concentrations of the above-mentioned nine constituents in different decoctions were simultaneously determined by HPLC and the linear equations of nine constituents were established. Compared with GZD, peoniflorin content increased in GLGZD significantly, while gallic acid, protoeateehuic acid, albiflorin, glycyrrhizin, liquiritin, cinnamic acid, glycyrrhizic acid content decreased in GLGZD significantly. Conclusion: This method had a good repeatability and reproducibility, which can be used for the quality control of GLGZD. Results also show that the contents change with GLGZD and GZD.
