摘要
探讨了固定化木瓜蛋白酶法脱除猫爪草粗多糖中蛋白质的工艺条件.以蛋白脱除率和多糖保留率为评价指标,采用正交试验研究了固定化酶用量、温度、pH、时间四个因素对脱蛋白效果的影响.结果表明最优工艺为:固定化酶用量12%,时间2.0 h,温度60℃,pH6.0,此条件下蛋白脱除率为73.2%,多糖保留率为71.3%.验证试验表明,此方法稳定可靠.
Removal of protein of Radix ranunculi crude polysaccharide by immobilized papain was investigated. With the removal rate of protein and retention polysaccharide as the index, four factors affecting the removal of protein including amount of immobilized enzyme,time,temperature,and p H were analysed using orthogonal test. The results showed that the optimal condition were as follows: amount of immobilized enzyme 12%, 2.0 h, 60 ℃, and p H6.0,removal rate of protein and retention rate of polysaccharide were 73.2% and 71.3%, respectively. Verification testing indicates that this method is stable and reliable.
