摘要
目的 探讨槲皮素治疗糖尿病肾病的机制。方法 对链脲佐菌素诱导糖尿病大鼠动物模型 ,给予槲皮素10 0 mg/ (kg· d) ,治疗共 8周。放免法测定尿白蛋白排泄量 ,免疫组化及 RT- PCR检测方法观察槲皮素对糖尿病大鼠肾脏皮质 TGF-β1 蛋白及基因表达的影响。结果 治疗 2周及 8周时 ,槲皮素治疗组糖尿病大鼠尿白蛋白排泄量明显下降 ,肾脏皮质 TGF-β1 免疫染色强度比未治疗组明显减轻 ,TGF-β1 m RNA水平比未治疗组明显下降 (P<0 .0 5 ,P<0 .0 1)。结论 槲皮素可抑制糖尿病大鼠肾脏皮质 TGF-β1 蛋白及基因过度表达 ,其防治糖尿病肾病的作用可能部分通过抑制TGF-β1 过度表达有关。
Objective To investigate the mechanism of Quercetin treatment on diabetic nephropathy.Methods Quercetin 100mg/(kg·d) was given to diabetic rats.Urinary albumin excretion was measured by radioimmunoassay.The expression of TGF β 1 protein and TGF β 1 mRNA in renal cortex of diabetic rats were determined by immunohistochemical staining and RT PCR analysis.Results Quercetin treatment decreased urinary albumin excretion in diabetic rats.TGF β 1 immunohistochemical staining and TGF β 1 mRNA levels in Quercetion treated group was significantly decreased than untreated DM group ( P <0.05, P <0.01).Conclusion Quercetin inhibits TGF β 1 protein and mRNA over expression in renal cortex of diabetic rats,suggesting that Quercetin might play a beneficial role in experimental diabetic nephropathy.Its mechanism may be partly by inhibiting TGF β 1 protein and mRNA over expression in kidney.
