姜黄素对大鼠糖尿病肾的实验研究 被引量:24

摘要 目的研究姜黄素对糖尿病大鼠肾脏的保护作用并探讨其保护作用的机制。方法用链脲佐菌素诱导糖尿病大鼠动物模型,给予姜黄素250 mg/(kg·d)治疗共6周。测定尿白蛋白排泄量、尿TXB2、Ccr,肾组织光镜和电镜检查了解组织形态,肾组织免疫组化检查观察姜黄素对糖尿病大鼠肾脏皮质TGF-β1表达影响。结果治疗6周后,姜黄素治疗组糖尿病大鼠的尿白蛋白和尿TXB2排泄量明显下降,Ccr亦下降。光镜下和电镜下系膜区基质增生明显减轻,肾脏皮质TGF-β1免疫染色强度比未治疗明显减轻(P<0.05)。结论姜黄素对糖尿病大鼠肾脏有保护作用,其机制与抑制糖尿病大鼠肾皮质TGF-β1蛋白过度表达,以及抑制花生四烯酸类生化途径有关。 Objective:To investigate the protective effect of curcumin on renal lesions in diabetic rats and the mechanism.Methods:Curcumin250mg/kg·d was given to streptozotocin-induced diabetic rats.After6weeks,urinary albumin excretion and thromboxaneB 2 and Ccr were measured.Glomerular structure and ul-trastructure of kidney were examined by light microscopy and electron microscopy.The expression of TGF-β 1 in renal cortex of diabetic rats were determined by immunohistochemical technique.Results:Curcumin de-creased urinary albumin,thromboxaneB 2 excretion and Ccr in diabetic rats.Compared with the untreated dia-betic rats,proliferation of mesangial matrix improved markedly.TGF-β 1 immunohistochemical staining in cur-cumin-treated group significantly decreased than that in untreated DM group(P<0.05).Conclusions:Cur-cumin can protect kidney of streptozotocin-induced diabetic rats.The mechanism may be partly mediated by inhibiting TGF-β 1 over-expression in kidney and inhibiting AA biochemical methods.
