摘要
目的 观察木黄酮 (genistein,GST)对培养的人垂体腺瘤细胞增殖的影响 .方法 将 GST作用于体外培养的人垂体腺瘤细胞 ,测定 MTT值及 3H- Td R参入量 ,并用流式细胞仪测定细胞周期 .结果 不同浓度的 GST(10 - 7,10 - 6 和10 - 5 mol· L- 1 )明显抑制垂体腺瘤细胞的增殖 ,MTT的A490 nm 值变化率分别为 (91.2± 4.9) % ,(78.8± 5 .3) %和(6 5 .8± 5 .6 ) % ,(P<0 .0 1) ,cpm值的变化率分别为 (88.6±3.6 ) % ,(75 .5± 5 .7) %和 (5 8.9± 4.9) % ,(P<0 .0 1) ,并存在着剂量效应 . 10 - 5 mol· L- 1 的 GST可使 G1期细胞比例从对照组的 6 8.2 %上升为 91.0 % (P<0 .0 1) .木黄酮对垂体腺瘤细胞的作用与雌激素受体无关 .结论
AIM To study the influence of genistein on the proliferation, DNA synthesis and cell cycle of cultured human pituitary tumors cells. METHODS MTT method and 3H TdR incorporation cell cycle analysis were used to examine the changes of proliferation and DNA synthesis of human pituitary tumors cells. RESULTS In a dose dependent manner, genistein at different concentration (10 -7 mol·L -1 , 10 -6 mol·L -1 , 10 -5 mol·L -1 ) could significantly inhibit the proliferation of human pituitary adenoma cells cultured in vitro . The change rate of A 490nm value were (91.2±4.9)%, (78.8±5.3)% and (65.8±5.6)% respectively ( P <0.01), and the change rates of cpm value were (88.6±3.6)%, (75 5±5.7)% and (58.9±4.9)% respectively ( P <0.01). GST (10 -5 mol·L -1 ) increased the proportion of cells in G 1 phase from 68.2% up to 91.0% ( P <0.01). Moreover, the effect of genistein on the proliferation of cultured human pituitary tumors cells was independent of estrogen receptor. CONCLUSION Genistein inhibits the proliferation, DNA synthesis and cell cycle of cultured human pituitary tumors cells, and its inhibitory effect is independent of estrogen receptor.
