摘要
为探讨水产常用抗寄生虫药物伊维菌素(IVM)对鱼类的毒性效应,参考IVM对斑马鱼的96h—LC50值,设定了7μg/L、14μg/L及21μg/L3个处理组,并以0.021%乙醇为试剂对照组,采用96h半静水式毒性试验测定了MM对斑马鱼(Dartiorer/o)头部乳酸脱氢酶(LDH)、乙酰胆碱酯酶(AChE)和碱性磷酸酶(ALP)以及肌肉组织中超氧化物歧化酶(SOD)、丙二醛(MDA)和ALP等生理生化指标的影响。结果表明:IVM对斑马鱼肌肉SOD活性的影响总体表现为低质量浓度抑制,高质量浓度先诱导后抑制;MDA含量在低质量浓度作用下8~12h内显著高于对照组(P≤0.05),而在较高质量浓度下先受到显著抑制(P〈0.05),虽然后期有一定的恢复,但总体仍低于对照组;IVM对AChE活性影响在短期内并不明显,但在96h时最高质量浓度组的1VM对AChE产生了55.2%的抑制;头部LDH活性只在8~24h内受到不同程度的影响,之后便逐渐恢复;头部ALP活性在较高质量浓度作用12—24h内受到一定的抑制(P〈0.05),而肌肉ALP活性与对照组相比无显著差异(P〉0.05),表明头部ALP活性比肌肉敏感。
Abstract : This paper is inclined to present the results of our study on the effects of ivermectin on the physiological and biochemical charac- teristic features of Danio rerio. As is known, ivennectin is a kind of toxic antiparasitc medicine widely used in aquaculture, which is badly needed to overcome the toxic effects of the medicine on the healthy growth of the above mentioned fish. To solve the problem under ques- tion, we have done three groups of experiments in different IVM con- centrations of 7 μg/L, 14μ/L, 21μg/L with a control group of 0.021% ethanol set up as a control group. During the 96-hours' static acute toxicity experiment, we have carefully observed and exam- ined the indices of the physiological and biochemical characteristics, such as the activities of lactate dehydrogenase ( LDH), acetylcholin esterase (ACHE), and the alkaline phosphatase (ALP) in the fish head. We have also detected the superoxide dismutase (SOD), the malonaldehyde (MDA) and ALP in the muscle of the fish. In com- parison with the control group, we have noticed the effects of IVM on SOD activity in Danio rerio muscle displaying in a trend of inhibition at the low concentration levels, in which the SOD activity indicates a trend of first increase and then decrease at the higher IVM concentra- tions. To be exact, we have noticed that during the first 12 hours, it has been noticed that the MDA content in the fish muscles of 7μg/L group proved significantly higher than that of the control group (p 〈 0.05 ), whereas the content of MDA in the muscles of the experimen- tal group (the group of 14μg/L and 21 μg/L) demonstrated signifi- cantly inhibitive, though there exists a trend of recovery, with the content remaining lower than that in the control group. Yet, on the whole, as compared with the control group, in the short period, no remarkable difference was found in the effect of IVM on the AChE ac- tivity. Nevertheless, the AChE activity of the fish head with the high- est concentration toxic experimental group (21 μg/L) proves quite se- riously inhibited as a result of the 96 h tests. The inhibition ratio turned out to be 55.2 %. With the LDH activity in their head fluctu- ating as a result of 8 h to 24 h' s experiment, though longer free from the toxic exposure a trend of recovery has been noticed. As to the ALP activity with the heads of the fish of the 14 μg/L and 21μg/L group, a significant inhibitive effect could be found after the process of 12 h to 24 h (p 〈 0.05) test. However, no much difference could be found when compared with the control group in the muscle (p 〉 0.05), which indicates that the activity of ALP in the head is sup- posed to be more critical and sensitive than that in their body mus- cles.
