摘要
文章采用固相萃取-高效液相-紫外(SPE-HPLC-UV)联用法同时检测土壤中磺胺类、四环素类和氯霉素类抗生素含量,对四川岷江上游流域养殖场土壤中残留抗生素进行风险评估。分时段采集6处不同养殖场土壤,采用乙腈-磷酸缓冲盐和EDTA提取液提取,使用SAX和HLB小柱固相萃取,C_(18)柱进行液相分析,草酸盐-乙腈为流动相,梯度洗脱,270 nm下紫外检测。根据结果,利用环境风险熵值法对上述地区不同时间环境土壤进行评估。结果表明:该检测方法中待测组分能达到有效分离且专属性强。在0.048~41.78μg/mL线性范围内,浓度与峰面积均呈良好线性关系(r>0.9995),不同种类的抗生素最低检出限范围为11~26μg/kg,磺胺类、四环素类和氯霉素类回收率分别为75.2%~79.8%、60.2%~63.0%和65.4%。不同时段土壤样品均检出不同种类抗生素,以四环素类含量最高,且冬季采样土壤中残留量大于春夏季。大部分抗生素残留的环境风险处于较低水平,但仍有部分地区处于中风险水平。该研究所建方法准确、快速、简便,可用于土壤中残留抗生素的检测,结合环境风险分析,有效指导土壤污染检测管理和降解评估。
To establish an SPE-HPLC-UV method for determining contents of sulfonamides,tetracyclines and chloramphenicols in soil,and evaluating the risk of residual antibiotics in livestock farm soil in Minjiang River valley in Sichuan Province,soil samples were collected at different times from six different livestock farms,and then the soil samples were extracted after adding with acetonitrile-phosphate buffer and EDTA.The extraction was purified and concentrated by SAX and HLB column.The column of C_(18)was adopted,and the oxalic acid buffer-acetonitrilel was used as mobile phase.The wavelength of detector was 270 nm.By analyzing the detection results,the environmental risk entropy method was used to evaluate the environmental soil in the above areas.Results showed that the ingredients to be determined can be separated effectively with strong specificity.Sulfonamides,tetracyclines and chloramphenicols had good linear relationships in the range of 0.048~41.78μg/mL(r>0.9995),the limit of detection was 11~26μg/kg,and the recoveries were 75.2%~79.8%,60.2%~63.0%and 65.4%.Different kinds of antibiotics were detected in those samples,and tetracycline was the highest.The environmental risk of most antibiotic residues is at a low level.This method is simple,rapid and sensitive for determination of residual antibiotics in soil,through environmental risk analysis,it can guide the management of soil pollution detection effectively.
