构建SD大鼠移植性胃癌模型及不同方法诱癌率的比较 被引量:9

Construction of implanted gastric cancer model of SD rats and comparison of inducing-cancer rates with different construction ways

摘要 目的采用瘤细胞悬液和瘤组织块原位种植构建Sprague Dawley(SD)大鼠原位移植胃癌模型,并比较其诱癌率。方法分别采用直接注射法和组织插块法构建SD大鼠胃癌模型:直接注射法组将walker-256细胞悬液直接注射到SD大鼠胃壁的浆膜内,观察移植肿瘤的生长状况、移植成功率;组织插块法组以walker-256细胞接种于SD幼鼠皮下,形成稳定传代的皮下移植瘤,再取该肿瘤组织块原位移植于SD大鼠胃壁,观察移植肿瘤的生长状况、移植成功率。结果直接注射法组原位移植成功率为5/7,造模9 d后平均瘤重为0.320 g;组织插块法组原位移植胃癌成功率为3/7,造模9 d后平均瘤重为0.206g。直接注射法组原位移植成功率和造模9 d后平均瘤重均较组织插块法组高。结论直接注射法原位移植模型的生物学行为与人胃癌自然生长过程相似,可作为一种有价值的工具用于胃癌转移机制和抗转移实验。 Objective To establish transplantation in situ gastric cancer model of Sprague Dawley (SD) rats with direct injection of tumor cell suspension (direct injection group) and orthotopic transplantation of tumor tis- sue block (tissue block insertion group) and compare inducing-cancer rates of two construction methods. Methods The gastric cancer models of SD rats were respectively established by the methods of direct injecting cell suspen- sion and tissue block insertion. In direct injection group,walker-256 cell suspension was directly injected into the se- rous membrane of gastric wall of adult SD rats, and the tumor growth conditions and the success rate of transplanta- tion were observed. In tissue block insertion group, the walker-256 cells were inoculated to subcutaneous tissue of immature SD rats, and the stable passage subcutaneous transplantation tumor was formed. Then the tumor tissue block was taken and in situ implanted into gastric wall of SD rats, and the tumor growth conditions and the success rate of transplantation were observed. Results In direct injection group, the success rate of transplantation in situ was 5/7, and the average tumor weight was 0.320 g at the ninth day after molding. In tissue block insertion group, the success rate of transplantation in situ was 3/7, and the average tumor weight was 0. 206g at the ninth day after molding. The success rate of transplantation in situ and the average tumor weight at the ninth day after molding in di- rect injection group were all higher than those in tissue block insertion group. Conclusions The biological behavior of SD rat orthotopic transplantation model with direct injection method is similar with natural growth process of human gastric cancer, and it can be served as a valuable tool for the experiments of transfer mechanism of gastric cancer and anti-transfer.
