摘要
从人胎脑cDNA文库中克隆到一种新的酪蛋白激酶的cDNA ,全长 175 4bp ,拟编码 438个氨基酸残基 ,该蛋白预测的分子质量为 5 0 2 72u ,等电点 9.37.经同源分析 ,该基因与大鼠的CK1γ1蛋白有 94%的相似性 ,为人 1型酪蛋白激酶 γ1亚型 (CK1γ1)基因 .人CK1γ1基因同时与 15号和 17号染色体的基因组部分序列完全一致 ,但RH法却将基因定位于 15 q2 2区段的D15S15 9 D15S12 5Marker之间 .多组织Northern膜 (MTN)杂交分析显示 ,人CK1γ1基因在肝、结肠、肾和骨胳肌特别在肝组织中有高表达 .
A cDNA coding for human protein kinase CK1(Casein Kinase 1) has been isolated by screening human 18 weeks fetal brain cDNA library and sequenced. This new cDNA of 1 754 bp contains an open reading frame of 438 amino acids whose deduced molecular mass is 50 272 u and isoelectronic point is 9.37. The entire amino acid sequence of novel human CK1 is 94% homologous to that of rat CK1γ1, and their sequence in the kinase domain is almost completely identical, predicting that novel cDNA is for a human CK1γ1 isoform. Tissue expression profile analysis have indicated that the human CK1γ1 is high expressed among the liver, skeletal muscle, heart and kidney. The human CK1γ1 gene was mapped to chromosome 15q22 between STS marker D15S159 and D15S125 by polymerase chain reaction analysis of human/rodent hybrid cell panels.
