摘要
将人酪蛋白激酶基因CK1γ1的cDNA与 40 96个胎脑cDNA一起点在玻璃介质的基因芯片上 ,用 16种不同组织或细胞系的mRNA分别杂交该基因芯片 ,以正常人混合组织mRNA作共同对照 ,检测CK1γ1基因的表达谱变化 ,结果显示人CK1γ1基因在 15种组织表达没有变化 ,但在肝癌组织中的表达却显著下调 .采集的 7例肝癌标本的cDNA与正常肝组织的cDNA重复 7次芯片杂交 ,结果肯定了CK1γ1基因在肝癌组织中表达下调 .同时 ,对芯片杂交结果通过聚类分析 ,按基因表达谱特征将CK1γ1基因与其他 7种基因聚类在一起 ,提示CK1γ1基因可能在生物学作用方面与这 7种基因有某种相关性 .
The cDNA microarray was used to analyze the expression profile of human CK 1 γ 1 cDNA cloned among 16 different tissues or cell lines. The result showed that human CK 1 γ 1 gene expressed at low level in hepatoma as compared with other tissues. 7 cases of repeated hybridization with array using cDNAs coming from hepatoma and normal liver respectively confirmed that the expression of human CK 1 γ 1 gene was specifically down regulated in hepatoma. Expression pattern based gene clustering has clustered CK 1 γ 1 with other seven genes, which suggests the possible functional co relationships among them.
