摘要
应用生物素一亲和素系统制备免疫磁珠,考察了不同条件对制备免疫磁珠的影响.结果表明:当反应温度为4℃、亲和素浓度为0.2 mg/mL、PBS pH=5.8、反应时间为2h时,磁性微球的亲和素载量为25~55 mg/g; BNHS与抗体质量比1:7时,生物素与抗体的结合率最高;亲和素与生物素化抗体摩尔比在1:3时,亲和素与生物素的偶联率最高.抗CD45免疫磁性微球的白细胞去除率高达78%,磁珠回收率达98%,可用于进一步捕获乳腺癌外周血循环肿瘤细胞.
The biotin avidin system was applied to prepare immuno-magnetic beads and investigate the effects of different conditions on the preparation of immuno-magnetic beads. The result showed that the neutravidin capacity of the immuno-magnetic beads was 25-55 mg/g when the synthetic reaction tempera- ture was at 4℃ ,density of neutravidin was 0.2mg/mL and 2h reaction in pH 5.8 of PBS. When BNHS and antibody mass ratio was at 1 : 7,the most antibody was bound to the biotin and when neutravidin and bio- tin molar ratio was at 1 : 3, the highest coupling ratio was get. The leukocyte removal ability of the anti- CD45 immune magnetic polymer microsphere was up to 78%. The recovery rate of microsphere was 98% and was able to be used to further capture of breast cancer tumor cells in the peripheral blood circulation.
