摘要
建立以微卫星不稳定性为指标,免疫磁珠-PCR技术检测尿中的肿瘤脱落细胞的方法。用膀胱癌单克隆抗体BDI-1与磁珠结合,制备出免疫磁珠BDI-1-IMB,与膀胱癌细胞有特异性吸附效应,而对人直肠癌细胞LOVO无此作用。结合PCR技术,建立了分选及检测血尿中的膀胱癌细胞株EJ细胞的方法。结果显示尿液中有5个EJ细胞即可被分选及检测出。该方法是一种特异强,敏感性高,简便快速的检测技术。
Division of Urology,The Third Hospital,Beijing Mecical University,100083 By employing microsatellite DNA marker,Immunomagnetic beads (IMB) and polymerase chain reaction (PCR),the method was developed to detect the droped tumor cells in urine.IMB against human bladder cancer cell were constructed by binding the monoclonal antibody BDI-1 with magnetic beads.BDI-1-IMB exerted the specific combining effect on human bladder tumor cells,but not on the human colon cancer cell line LOVO.Combined PCR technique,the mothod of sparation and detection of bladder cells line EJ in hematuria was established.The results showed the sensitivity of detection reached five cells. IMB-PCR is high specific,sensitive and rapid analysis technique and offer the tool for early clinical diagnosis of bladder cancer.
