摘要
目的 :探讨苦瓜蛋白抗肿瘤活性的机制。方法 :从新鲜的苦瓜种子中分离纯化苦瓜蛋白 ,通过MTT比色法分析其对SGC7901细胞以及正常人二倍体细胞2BS增殖的影响 ,并应用光镜、电镜观察及流式细胞术等手段 ,研究苦瓜蛋白诱发胃癌细胞SGC7901的形态学、DNA含量变化。结果 :苦瓜蛋白对胃癌细胞SGC7901的IC50约为18μg/ml,在相同剂量下对2BS细胞的抑制率仅为8 % ;与对照组细胞相比 ,加药后的SGC7901细胞发生典型的凋亡形态学改变及DNA含量变化。结论 :苦瓜蛋白对胃癌细胞SGC7901的增长具有明显的抑制作用 。
Objective:To study the antitumer mechanism of Momordin.Methods:SGC7901 cells were treated with Momordin which had been isolated and purified from seeds of the Momordica Caranata.The chemosensitivities of SGC7901 and normal diploid cell 2BS were examined with MTT assay in vitro.The morphological changes and DNA contents were detected by light microscopy,electronic microscopy and FACS assay.Results:The IC50 for SGC7901 is about 18μg/ml,while the lethal rate of 2BS is only about 8% at the same dose.Comparing with control cells,SGC7901 cells after the treatment with Momordin showed a typical apoptosis changes in morphology and DNA contents.Conclusion:Momordin could significantly inhibit the reproduction and induce apoptosis of SGC7901 cell.
