摘要
目的观察补中益气汤对脾气虚证大鼠手术切口愈合的影响。方法将60只SD大鼠随机分为正常组、脾气虚模型组、补中益气汤组,每组20只。除正常组外其余各组采用苦寒泻下法制备脾气虚动物模型,各组造模第19d于背部做长约4cm并平行脊柱的切口,补中益气汤组手术当天大鼠每日灌服7g/kg的补中益气汤水煎剂;脾气虚模型组和正常组大鼠每日给予等体积蒸馏水灌胃。各组分别于术后3d、7d各处死10只大鼠,取距手术切口两侧肌肉组织行HE染色,光镜下观察手术切口处炎症反应和肉芽组织增生情况,高倍镜下对切口肌肉组织成纤维细胞进行计数。结果补中益气汤组术后7d切口处可见新生肉芽组织,其间有大量新生血管,并有巨噬细胞、淋巴细胞浸润。与正常组术后3d、7d比较,脾气虚模型组成纤维细胞数量显著降低(P<0.01);与模型组术后3d、7d比较,补中益气汤组术后同时间点的成纤维细胞数量显著升高(P<0.05或P<0.01)。结论补中益气汤通过改善手术切口局部肌肉组织炎症反应和肉芽组织增生情况,可促进切口愈合。
Objective To observe the effect of Decoction, 补中益气汤） on incision healing in rats were randomized into the normal group, model group models of spleen qi deficiency pattern were established Buzhong Yiqi Decoction （Center-Supplementing Qi-Boosting with spleen qi deficiency pattern. Methods Sixty SD rats and Buzhong Yiqi Decoction group, with 20 in each. The rat in the model group and Buzhong Yiqi Decoction group. A 4 cm incision paralleled to the spine was made on the back of rats in the each of groups on the 19th day of modeling. The Buzhong Yiqi Decoction group was gavaged with 7g/kg of Buzhong Yiqi Decoction daily after operation. The normal group and model group were gavaged with an equal volume of distilled water. Ten rats were sacrificed respectively on the 3rd day and 7th day after operation in each group. The muscle tissue on both sides of surgical incision was sam- pled and dyed with routine HE staining. The inflammation and granulation tissue formation was observed. The fibro- blast cells were counted. Results The new granulation tissue with new blood vessels, macrophages and lympho- cytes could be seen in the Buzhong Yiqi Decoction group 7 days after operation. Comparing with the normal group, the number of fibroblasts was significantly decreased in the model group 3 days and 7 days after operation （P 〈 0. 01 ）. Comparing with the model group, the number of fibroblasts was significantly increased in the Buzhong Yiqi Decoction group 3 days and 7 days after operation （P 〈 0. 05 or P 〈 0.01 ）. Conclusion Buzhong Yiqi Decoction can promote incision healing by relieving inflammation and granulation tissue formation of muscle tissue.
