刺参酸性粘多糖对二乙基亚硝胺诱发肝癌大鼠细胞周期的影响 被引量:1

EFFECT OF STICHOPUS JAPONICUS ACIDIC MUCOPOLYSACCHARIDE ON THE CELL CYCLE IN RAT LIVER CANCER INDUCED BY DIETHYL NITROSAMINE

摘要 目的观察刺参酸性粘多糖(SJAMP)对二乙基亚硝胺(DEN)诱发大鼠肝癌形成的抑制作用并研究其对细胞周期的影响。方法大鼠灌胃DEN诱癌,同时灌胃不同浓度sJAMP,16w后停药,处死大鼠。观察大鼠肝脏结节发生情况及肝细胞形态;RT-PCR法检测细胞周期素(cyclinD1)、细胞周期蛋白依赖激酶(CDK4mRNA)的表达;Western-blot法检测pRb(retinoblastoma protein)、E2F-1(adenovirus E2 factor-1)的表达。结果sJAMP各干预组结节数均低于模型组。电镜下发现,sjAMP干预组的大鼠肝细胞中出现数量不等的凋亡小体。RT-PCR结果显示,各组大鼠cyclinDl表达降低(F=49.91,q=5.94~18.52,P〈0.05);CDK4表达降低(F=50.21,q=5.72~18.08,P〈0.05)。Western-blot结果显示,各组大鼠pRb蛋白表达增加(F=63.25,q=3.56~16.36,P〈0.05);E2F-1蛋白表达降低(F=268.10,q=14.55~35.45,P〈0.05)。结论sjAMP对DEN诱发大鼠肝癌形成有抑制作用,其机制可能通过抑制cyclinD1、CDK4mRNA、E2F-1的表达,诱导细胞周期阻滞,从而抑制肝癌细胞过度增殖。 Objective To observe the effects of Stichopus japonicus acidic mucopolysaccharide (SJAMP) on the cell cycle in liver cancer induced by diethyl nitrosamine (DEN). Methods Rats were fed DEN to induce cancer, and treated with different doses of SJAMP. The rats was killed and the liver morphology was observed by an electron microscopy The cyclinD1 and CDK4 mRNA expressions were detected by RT-PCR. pRb (retinoblastoma protein) and E2F-1 (adenovirus E2 factor-l) were detected by Western blotting. Results The number of liver cancer nodules in SJAMP groups was less than those in model group. Apoptotic bodies were found in rat hepatocytes in SJAMP groups under electron microscopy. CyclinDland CDK4 expressions in SJAMP groups were significantly lower than those in model group (P〈0.05). Western blotting results showed that compared with the model group, the pRb expression in other groups was increased (F=63.25,q=3.56-16.36,P〈0.05); E2F-1 expression decreased(F=268.10, q=14.55-35.45, P〈0.05).Conclusion SJAMP can inhibit the formation of liver cancer induced by DEN. SJAMP may inhibit cyclinD1, CDK4 mRNA and E2F-1 expressions, induce cell cycle arrest and inhibit cancer cell proliferation. [ACTA NUTRIMENTA SINICA, 2013,35(6): 582-586]
