摘要
以转EGFP-attB基因PK15细胞(猪肾细胞15)为试验材料,研究不同秋水仙素浓度、低渗时间对染色体标本制备的影响。结果表明,使用秋水仙素浓度为0.04~0.08μg/mL、低渗时间为15~20 min时所制备的染色体标本分裂相分散好,可以做核型分析用,为下一步做染色体荧光原位杂交检测研究转EGFPattB基因在染色体上的整合位点奠定了基础。
The porcine kidney cells 15 transfected with EGFP-attB gene were used to prepare chromosome specimen. The re- sults showed that the final concentration of 0.04~0.08 μg / mL colchicine and 15~20 min of low hypotonic treatment was the best parameter. The mitosis of chromosome specimen is suitable for karyotype analysis, and provides basis for using chromo- some fluorescence in situ gybridization to study the integration site of porcine kidney cells 15 transfected with EGFP-attB gene.
