Transcription activator effector nucleases (TALENs) is a novel DNA editing technology which can induce precise site-specific modification in genome. TALENs consist of two distinct domains, an engineered DNA-binding domain derived from the transcription activator-like effector (TALE) that provides the capability and specificity for binding target DNA sequence, and a non-specific restriction endonuclease Fok I domain fused to the C-terminal of TALE that confers the nuclease activity of TALENs. Here, we summarize the technological back- ground and provide an overview of development and applications of TALENs in genome engineering.
