摘要
目的 观察铁缺乏对大鼠耳蜗肌动蛋白的影响 ,探讨铁缺乏致聋的发病机制 .方法 应用 SDS- PAGE蛋白电泳、双波长薄层扫描分析、免疫组织化学及听性脑干诱发电位检测 ,对照观察 6~ 8wk铁缺乏大鼠和正常大鼠耳蜗肌动蛋白含量及分布的变化与听阈变化的关系 .结果 铁缺乏引起感音神经性聋大鼠耳蜗肌动蛋白相对含量及免疫反应活性均有明显下降 .结论 耳蜗内肌动蛋白含量减少和 (或 )免疫反应活性降低是铁缺乏致聋的重要病理基础之一 .
AIM To observe the effect of iron deficiency(ID) on cochlear actin in rats.METHODS Actin of cochlear membranous tissue was separated and determined by SDS-PAGE and Western blot; the relative content and distribution of cochlear actin in both normal and iron deficient rats were studied by TLC-densitometric determination and immunohisto- chemistry. RESULTS 37.5%(18/48 ears)ID rats had substantial elevation of ABR hearing threshold (>15 dB SPL) and reduction of relative content and immunohistical reaction of cochlear actin. CONCLUSION Reduction of actin in cochlea is one of the important pathological bases for sensorineura hearing loss in iron-deficient rats.
