摘要
目的对鼻息肉家系进行全基因组扫描,从而找出鼻息肉易感基因的位点。方法收集了8个鼻息肉家系,46份外周血标本,其中鼻息肉患者26例,健康者20例,以800个分布在23条染色体上的荧光标记的微卫星DNA序列为引物,进行基因扫描和基因定型,并将结果进行连锁分析。结果8个家系全基因组扫描未能发现存在易感基因位点的区域。结论虽然本实验未能找到鼻息肉易感基因位点,其原因可能与所收集的标本量不够有关,最后结果的确定需更多的鼻息肉家系来支持。
Objective To identify nasal polyp susceptibility gene locus by genome-widp scan for familial nasal polyps. Methods 46 blood samples were obtained from 8 nasal polyps families. There were 26 patients and 20 healthy,althy controls. DNA fragments were amplified by PCR. 800 fluorescence-labeled markers were used for genome-wide scan. Genotyping and linkage analysis were performed, Result No susceptibility gene locus was found in the 8 families by linkage analysis. Conclusion Although no susceptibility gene locus is found in this study, more nasal polyps families are needed for a definite conclusion.
