摘要
以草莓灰葡萄孢霉、辣椒炭疽病菌、桃褐腐病菌、棉花红腐病菌、马铃薯干腐病菌植物病原真菌为试材,采用适于大规模分离抑菌活性物质的大孔树脂等分离技术从瑞香狼毒根中分离抑菌活性物质,并确定了瑞香狼毒根部抑菌活性物质提取工艺。结果表明:瑞香狼毒根宜用乙醇浸提法获得粗提浸膏,再用乙酸乙酯进一步萃取,随后用大孔树脂进一步分离,得到的组分9对灰葡萄孢霉等有较好的抑菌效果(半抑制浓度(IC50)值为0.39mg/mL)。核磁鉴定组分9的主成分为狼毒色原酮,抑菌活性测定结果表明狼毒色原酮对草莓灰葡萄孢霉、桃褐腐病菌和辣椒炭疽病菌等均有较好的抑菌效果。
Taking Botrytis cinerea Pers. , Colletotrichum acutatum, Monilinia fructicola, Fusarium monili forme and Pythium coeruleum as test fungi, screening antifungal compounds from the root crude extracts of SteUera chamaejasrne, and separated the major antifungal compounds from Stellera charnaejasrne through adapting the macroporous resin, which was suitable for the large-scale separation, the antifungal compounds extraction process were established. The results showed that the ethanol extraction followed by ethyl acetate partition, could better isolate antifungal compounds from root of S. chamaejasme. The antifungal components of ethyl acetate extract of S. chamaejasrne were further efficiently partitioned using the macroporous resin. The 9th elution fraction eluted from macroporous resin column was found to have highest antifungal activity against B. cinerea with an IC40 value of 0. 39 mg/mL. The main component of the 9th fraction of S. chamaejasme was separated and identified as chamaechromone using HPLC and NMR. Chamaechromone was first found to be able to effectively inhibit the growth of mycelium of B. cinerea, C. acutaturn, and M. fructicola.
