摘要 目的寻找一种更好的测定肌苷的方法。方法以固定化核苷磷酸化酶(EC 2.4.2.1)、黄嘌呤氧化酶(EC 1.2.3.2)与过氧化氢电极构成电流型酶电极,研究了肌苷酶电极分析法的各种分析条件及参数,并与柱层析法对比测定了肌苷发酵液中肌苷含量。结果酶电极法测定结果与柱层析法测定结果之间有较好的一致性(r=0.9448;F=8.848,F0.01(2,16)=19.44)。结论采用酶电极法测定肌苷发酵液中肌苷含量具有专一性高、成本低、分析速度快等优点。 Objective To search for a more better dectection method used to inosine level. Methods Nueleoside phosporylase and xanthine oxidase was co-immobilized at a hydrogen peroxide electrode, the enzyme electrode was used for the amperometrie determination of inosine in its fermentation. Results The corrlation coefficient of the results between enzyme electrode and column chromatography method was 0.8844 which had better accordance (r=0.9448;F=8.848,Fo.ot (2,16)=19.44). Conclusion Inosine in fermentation could be determined specially, rapidly and inexpensively with enzyme electrode.
