摘要
以枯草杆菌(Bacillussubtilis)TX2(遗传标记Xan~-)为供体,以枯草杆菌TSA19(遗传标记Ade~-+His~-+8-AG~r+6-MP~r+SG~r)为受体,利用转化法成功地获得肌苷产量高、遗传稳定性好的目的转化子TSXB29。采用均匀设计理论,利用微机对实验数据进行统计处理,找出了该转化子的最佳摇瓶发酵条件,在该条件下,平均产苷为27.85g/L。经连续摇瓶和斜面传代,发现该转化子是稳定的,具有一定应用价值。
Bacillus subtilis TX2(Xan^-)being used as the donor,Bacillus subtilis TSA19(Ade^-+His^-+8-AG^r+6-MP^r +SG^r) as the recipient, with the method of transformation, a stable higher inosine-productivity transformant TSXB29 was obtained successfully. Using the theory of Uniform Design, the experimented data were analysed with the computer, then the optimum flaskshaking fermentable conditions were determined. under the optimum conditions, the transformant accumulates an average of 27.85g/l of inosine. After several generations in the shaking flask and several generations on the slope,the transformant was found stable,so it has the value of application.
