摘要
目的 探讨神经细胞凋亡与缺氧和机械损伤的关系。 方法 体外原代培养大鼠神经元细胞 ,经低氧或机械损伤处理后 ,应用光镜、电镜、末端脱氧核苷酸介导的X -dUTP缺口末端标记 (TdT -mediateddUTPneikendlabeling ,TUNEL)法、流式细胞仪、DNA凝胶电泳及免疫组化分析细胞凋亡及其相关基因表达的改变。 结果 经低氧或机械损伤后神经细胞出现凋亡的形态改变 ;其DNA呈现“梯状”断裂现象 ;细胞凋亡百分率分别为 10 .8%和 4.8% (对照组为 1.6 % ) ,凋亡相关基因 (c-myc、fas、fasL)表达明显增加。结论 缺氧及机械损伤可诱导神经细胞凋亡 ,其分子机制可能与某些凋亡相关基因表达改变有关。
Objective To explore the relationship between hypoxia or traumatic injury and neuronal apoptosis. Methods Rat neurons primarily cultured in vitro were made to be with hypoxia or traumatic injury. The neuronal apoptosis was evaluated with microscope, TUNEL staining, flow cytometry, agarose gel electrophoresis and immunohistochemistry. Results Morphological changes of apoptosis appeared in the treated neurons. DNA fragmentation showed a ladder break. The apoptosis ratio was 10.8% in hypoxia group and 4.8% in trauma group, while it was only 1.6% in the control group. The expression of apoptotic associate genes (c-myc, fas and fasL) was increased. Conclusions Hypoxia or traumatic injury can induce neuronal apoptosis; and its molecular mechanism is possibly related to expression of apoptotic associate genes.
