摘要
目的对一起由多种血清型副溶血性弧菌引起的食物中毒病原菌进行毒力基因及分子分型研究。方法参照国家标准方法对事件中可疑样品进行病原菌分离鉴定,对分离的副溶血性弧菌进行血清学试验,采用荧光定量PCR检测菌株的毒力基因耐热直接溶血素基因(tdh)、耐热直接溶血素相关溶血素基因(trh)与不耐热溶血素基因(tlh),并用脉冲场凝胶电泳(PFGE)对分离到的菌株进行分子分型。结果从24份病人标本和1份剩余食品中检出副溶血性弧菌25株;血清型分布为O4:K8 4株,O3:K6 19株,O4:KUT 1株和O3:KUT 1株;分离到的25株副溶血性弧菌毒力基因检测均为tdh+tlh+trh-;PFGE聚类分析显示,不同血清型菌株分为不同的簇,同一血清型菌株的PFGE条带型间存在1~2个条带的差异,菌株间存在关联。结论这是一起由多种血清型副溶血性弧菌引起的食物中毒。
Objective To investigate virulence genes and molecular typing for pathogens from a food poisoning case caused by multiple serotypes of Vibrio parahaemolyticus.Methods Foodborne pathogen from those suspicious foodborne poisoning samples were isolated and identified.Vibrio parahaemolyticus isolated was tested by serological experiments.The strain’s virulence genes tdh(thermostable direct hemolysin),trh(tdh related hemolysin),and tlh(thermolabile emolysin) were analyzed by real-time PCR,and molecular typing was performed on the isolated strains using pulsed-field gel electrophoresis(PFGE).Results In this study,25 strains of Vibrio parahaemolyticus were obtained from 24 patients and one left-over food.The distribution of serotype included 4 strains of O4:K8,19 strains of O3:K6,1 strain of O4:KUT,and 1 strain of O3:KUT.The virulence genes for the 25 isolated strains of Vibrio parahaemolyticus were all identified as tdh+tlh+trh-;PFGE analysis indicated that different serotypes were divided into different clusters,while the difference of 1 to 2 strips existed between the PFGE strips for the same serotype strains,and correlation was found among strains.Conclusions This is a food poisoning case caused by multiple serotypes of Vibrio parahaemolyticus.
