枸杞黄酮类化合物色谱分析条件的优化 被引量:6

摘要 目的优化枸杞黄酮类化合物的液相色谱分析条件。方法采用反相高效液相色谱法对枸杞黄酮类化合物的检测波长、流动相、洗脱方式分析条件进行优化。结果选择岛津VP-ODS 250 mm×4.6 mm(5μm)色谱柱;流动相A:乙腈∶水∶乙酸(5∶29.5∶0.5),流动相B:乙腈∶水∶乙酸(70∶94.5∶0.5);流速0.6 mL.min-1,梯度洗脱(0~14 min,100%~86%A;14~40 min,86%~70%A;40~55 min,70%~100%A;55~60 min,100%~0%A),检测波长342 nm,柱温30℃,进样量5μl。方法重现性好(芦丁峰面积RSD 2.70%,绿原酸峰面积RSD2.53%)。结论采用乙腈体系为枸杞黄酮类化合物色谱分析的流动相、梯度洗脱、黄酮类化合物各组分保留时间适中,分离效果好,可为高效液相色谱法检测枸杞黄酮类化合物提供一定参考。 Objective To improve the chromatographic analysis conditions of flavonoids in Lycium barbarum.L.Methods The reversed phase high performance liquid chromatography was applied to improve the analysis condition including mobile phase,detection wavelength and elution modes.Results The separation was performed on a Shim-pack VP-ODS 250 mm × 4.6 mm(5 um),acetonitrile / water / acetic acid(5∶ 94.5∶ 0.5) was used as the mobile phase A and acetonitrile / water / acetic acid(70∶ 29.5∶ 0.5) as the mobile phase B with gradient elution(0 ~ 14min,100% ~ 86% A;14 ~ 40min,86% ~ 70% A;40 ~ 55min,70% ~ 100% A;55 ~ 60min,100% ~ 0% A),the flow rate was 0.5 ml.min-1,the detective wavelength was at 342 nm and the temperature was 30 ℃.The method had good reproducibility(the RSD of Rutin peak area was 2.70% and Chlorogenic acid peak area was 2.53%).Conclusion Using acetonitrile system as chromatographic mobile phase and gradient elution,and he retention time of flavonoids component is moderate,and good separation effect,the improved method can provide certain reference by using high performance liquid chromatographic to detect the flavonoids in Lycium barbarum L.
