高效液相色谱法测定不同产地宁夏枸杞中芦丁和绿原酸的含量 被引量:15

摘要 目的建立反相高效液相色谱法同时测定宁夏枸杞中芦丁和绿原酸含量的方法。方法色谱分析条件Shim-packVP-ODS色谱柱(250 mm×4.6 mm,5μm),以乙腈-水-乙酸(5∶94.5∶0.5)为流动相A,乙腈∶水∶乙酸(70∶29.5∶0.5)为流动相B,采用梯度洗脱,流速0.5 ml·min^-1,检测波长342 nm,柱温30℃。结果绿原酸和芦丁线性范围均为10~100μg·ml^-1(相关系数分别为r=0.999 5和r=0.999 8),平均回收率(n=5)分别为98.6%,99.0%;不同产地、不同年份宁夏枸杞样品中芦丁和绿原酸含量存在差异。结论该方法重现性好,适用于同时测定枸杞中芦丁和绿原酸的含量。 Objective To develop an RP-HPLC method with gradient dilution for simultaneous determination of chlorogenic acid and rutin in Lycium barbarum L from different habitats. Methods The separation was performed on a Shim - pack VP-ODS 250 mm×4.6 mm (5μm), acetonitrile/water/acetic acid(5: 94.5: 0.5)was used as the mobile phase A and acetonitrile/water/ acetic acid(70: 29.5: 0.5)as the mobile phase B with gradient elution, the flow rate was 0.5ml·min^-1 ,the detection wavelength was at 342 nm and the temperature was 30℃. Results The good linearities of chlorogenic acid and rutin were obtained over the ranges of 10 ~100μg·ml^ -1 ( r = 0. 999 5 and r = 0. 999 8 respectively) , the average recoveries were 98.6% and 99.0% respectively(n = 5). The contents of chlorogenic acid and rutin in Lycium barbarum L, from different habitats were different. Conclusion The method is rapid and reproducible, the method can be used to determine chlorogenic acid and rutin in Lycium barbarum L, from different habitats.
