老年和青年豚鼠喉部水通道蛋白1~5表达的研究

摘要 目的探讨青年和老年豚鼠喉部水通道蛋白1~5(aquqporin,AQP1~5)的表达及意义。方法选取健康4月龄豚鼠(青年组)和2年龄豚鼠(老年组)各10只,用免疫组织化学染色法观察两组豚鼠喉部组织AQP1~5的表达,用Image-Pro Plus 6.0For Window图像分析软件处理,比较两组结果。结果两组豚鼠喉部组织均有AQP1~5表达,AQP1主要表达在喉黏膜假复层纤毛柱状上皮、固有层的腺体以及小血管内皮细胞;AQP2主要表达于黏膜固有层的粘液性腺体和炎性浸润细胞,黏膜上皮细胞无或弱表达;AQP3高表达于黏膜假复层纤毛柱状上皮,腺体、血管内皮细胞和肌纤维组织均无表达;AQP4主要表达于喉黏膜鳞状上皮、假复层柱状纤毛上皮、固有层黏膜腺体、肌纤维及部分浸润细胞,血管内皮细胞无表达;AQP5主要表达于喉黏膜假复层纤毛柱状上皮细胞、上皮基底细胞、黏膜下浆液性腺体及导管,粘液性腺体无表达。青年组喉部AQP1、4的表达高于老年组,AQP5的表达低于老年组(P<0.05),两组间AQP2、3的表达差异无统计学意义(P>0.05)。结论豚鼠喉部组织有AQP1~5表达,老年豚鼠AQP1、4表达降低、AQP5表达升高的变化可能与其喉部老化改变有关。 Objective To study the expression of aquaporin(AQP)1~5in the larynx of young and old guinea pig.Methods Ten young(4months old)guinea pigs and 10old(2years old)guinea pigs were included in this study.The expression of AQP1~5in the larynx of guinea pigs were detected using immunohistochemistry methods.Results AQP1was expressed on the pseudo multiple cilium columnar epithelium,glands of the lamina propria and endotheliocyte of the micro vessel under the epidermis.AQP2was detected on the glands of the lamina propria mucosa,AQP3was observed on the pseudo multiple cilium columnar epithelium.AQP4was expressed on the squamous epithelium,the pseudo multiple cilium columnar epithelium,the glands of the lamina propria mucosa and the muscular cells.AQP5was expressed on the the pseudo multiple cilium columnar epithelium,basal cells,serous glands and pipes.The expression level of AQP1and AQP4in the young group was higher than that in the old group,while the expression level of the AQP5was lower(P0.05).The expression level of AQP2and AQP3showed no significant difference between the two groups(P0.05).Conclusion AQP1~5were expressed in the larynx of the guinea pig.AQP1,AQP4and AQP5may be associated with the aging of the guinea pig larynx.
