摘要
本文研究了维甲酸(RA)对体外培养的新生大鼠皮肤上皮基底细胞DNA和蛋白质合成的影响。RA(16 μmol/L)明显降低基底细胞~3H-dT掺入;显著增加~3H-亮氨酸和~3H-组氨酸掺入细胞蛋白质。经聚丙烯酰胺凝胶电泳与双相电泳辅以放射自显影术,证实有5种分子量不同的蛋白质受RA诱导合成,其中3种蛋白质为细胞表面膜蛋白。用~3H-甘露糖标记细胞糖蛋白糖链,发现RA可促进~3H-甘露糖的掺入,而且细胞表面糖蛋白中~3H-甘露糖掺入量比对照组增加43%。结果提示RA抑制上皮基底细胞增殖,诱导细胞合成蛋白质,主要为蛋白质的转换率增强,以及增加糖蛋白糖链合成。
The effect of retooio acid (EA) on DNA and protein synthesis in primary cultured basal cell from newborn rat was studied. BA (16μmol/L) decreased the 3H-dT incorporation into DNA of cells and increased the 3H-Leu, H-His into proteins. It was found that 5 proteins of various molecular weights were induced by RA. Three of them were surface membrane associated, proteins, determined by polyaorylamide gel eleotrophoresis and two dimensional electrophoresis with autoradiography. RA can stimulate 8H-mannose incorporation into cell glycoproteiins. There is a 43% increase as compared to control cells. These results suggested that RA inhibits the-proliferation of culture cells; induces protein synthesis, mainly with the increasing protein turnover and stimulates synthesis of the carbohydrate chain of N-linked glyooproteins.
