肉桂醛与盐酸小檗碱联合氟康唑抗白念珠菌作用的体外实验观察 被引量:6

摘要 目的探讨肉桂醛、盐酸小檗碱分别单独及联合氟康唑抗白念珠菌的作用及机制。方法采用体外药物敏感试验方法检测肉桂醛、盐酸小檗碱单独及联合氟康唑对白念珠菌的抗菌活性;通过逆转录聚合酶链式反应技术及凝胶分析软件检测和分析药物对白念珠菌PLB1基因的mRNA表达影响。结果肉桂醛、盐酸小檗碱单独对白念珠菌具有明显抑制其生长的作用,与氟康唑联合时,均具有协同作用;药物处理的白念珠菌PLB1的mRNA表达水平有明显差异,联合时比单用时其表达水平明显降低。结论肉桂醛、盐酸小檗碱、氟康唑单独及联合使用都具有明显的抗白念珠菌作用,药物联合时,白念珠菌的PLB1基因mRNA的相对表达量明显低于药物单用。 Objective To investigate the inhibitory effects and mechanisms of cinnamic aldehyde (CIN), berberine hydrochloride (BER) alone or combined with fluconazole (FLC) against Candida al- bicans. Methods The inhibitory effects of CIN, BER alone or combined with FLC against Candida albi- cans were detected through the in vitro susceptibility test The Candida alhicans PLB1 mRNA expression was measured by reverse transcription polymerase chain reaction ( RT - PCR) and analyzed by Gel - Pro analysis software. Results CIN and BER had obviously inhibitory effects on the growth of Candida albi- cans, respectively, where CIN and BER combined with FLC had a synergistic effects. There was signifi- cant difference of the Candida alhicans PLB1 mRNA expression among CIN, BER and FLC. The PLB1 mRNA expression after the combination of CIN, BER and FLC treatment was lower than that after single drug treatment alone. Conclusion CIN and BER alone or combined with FLC have apparently inhibitory effects against Candida albicans. The Candida albicans PLB1 mRNA expression after CIN and BER combined with FLC treatment was lower than that after single drug treatment alone.
