摘要
目的 :观察重组人白细胞介素 11(recombinanthumanIL 11,rhIL 11)对照射小鼠造血恢复的影响。方法 :给 5.5Gy60 Coγ射线照射小鼠连续 10d皮下注射rhIL 112 0 0 ,4 0 0 μg/ (kg·d)或溶剂 ,观察体重、外周血象和骨髓造血祖细胞的变化。结果 :各给药组小鼠体重明显增加 (P <0 .0 1)。照后第 11天rhIL 112 0 0 μg/ (kg·d)组血小板计数检查值明显高于照射对照组 (P <0 .0 5) ,各给药组第 2 0天检查值已高于照前值 ( 10 5%~ 112 % ) ,而照射对照组为照前值的 81%。骨髓有核细胞培养结果表明 ,各给药组的各系造血祖细胞集落数均明显高于照射对照组。结论 :rhIL 11可明显促进γ射线照射小鼠骨髓有核细胞形成CFU MK和CFU Mix ,升高外周血小板数。
Objective:To observe the effects of recombinant human IL-11(rhIL-11) on recovery of progenitor cells of bone marrow and peripheral platelets of γ-irradiated mice. Methods: RhIL-11 200 μg/(kg·d) or 400 μg/(kg·d), sc was administered to mice on days 0-9 after TBI with 5.5 Gy 60 Co γ-irradiation. The changes in body weight, peripheral blood cell counts and bone marrow hematopoietic progenitor cells of the mice were determined. Results: Animals administered with rhIL-11 gained more weight than the controls did. Daily administration of rhIL-11 promoted platelet recovery, resulting in less profound nadirs and a rapid recovery to normal levels. rhIL-11 treatment did not influence leukocytes. More colony forming units were found in IL-11 treated mice, especially CFU-MK and CFU-Mix. Conclusions: These results show that rhIL-11 can augment the platelet counts and strongly promote the recovery of megakaryocytic progenitor cells of bone marrow in mice irradiated with sublethal dose of 60 Co γ-rays.
