泥胡菜总黄酮大孔树脂纯化工艺优化 被引量:2

Optimization of Purification Technology of Total Flavonoids from Hemisteptia lyrata by Macroporous Resin

摘要 目的:优选大孔树脂纯化泥胡菜总黄酮的工艺条件。方法:采用静态吸附-洗脱试验筛选大孔树脂型号,动态吸附法优化大孔树脂纯化泥胡菜总黄酮工艺参数。结果:优选的纯化工艺为选用D101型树脂,上样液质量浓度1.5 g.L-1,上样速率3 BV.h-1,上样体积4 BV,上样液pH 5,加50%乙醇5 BV以2 BV.h-1流速洗脱,泥胡菜总黄酮纯度达47.8%。结论:D101型大孔树脂对泥胡菜总黄酮具有良好的纯化效果。 Objective: To optimize purification technology conditions of total flavonoids from Hemisteptia lyrata by macroporous resin.Method: Two models of macroporous resins were selected by static adsorption-elution test,purification technology of total flavonoids from H.lyrata with macroporous resin was optimized by dynamic adsorption method.Result: D101 resin was adopted,optimized purification technology was as following: the concentration of sample solution 1.5 g.L-1 with pH 5,adsorption velocity 3 BV.h-1,sample volume 4 BV,then eluted with 5 BV 50% ethanol at the speed of 2 BV.h-1,collected eluate.Under these conditions,yield of total flavonoids was 47.8%.Conclusion: D101 macroporous resin could purify total flavonoids from H.lyrata effectively.
